ANDARINE (S4)
S-4 is a SARM more commonly known as Andarine, also has been called Androxolutamide and GTx-007. In a fascinating study conducted by Gao et. Al, S-4 was characterized as an androgen receptor agonist in orchidectomized rats, with the mechanism of action in the skeletal muscle, bones, and pituitary.
S-4 Effects on Muscle Mass
Male rats were treated with either 3 mg/kg or 10 mg/kg of S-4 12 weeks after castration over an 8 week period. The study showed that both the 3 mg/kg dose and 10 mg/kg dose were successful in restoring both muscle mass and strength in the soleus muscle as well as muscle mass in the levator ani muscle comparable to that of intact male rats (https://www.ncbi.nlm.nih.gov/
Treatment with S-4 also minimally impacted the weight of the prostate and the seminal vesicles by approximately 16-17%, however, the S-4 treatment had strong androgenic effects on the prostate and seminal vesicles.
Furthermore, both the 3 mg/kg dose and the 10 mg/kg dose demonstrated strong agonist activity in the pituitary by decreasing plasma levels of both luteinizing hormone (LH) and follicle-stimulating hormone (LSH) in the castrated male rats (https://www.ncbi.nlm.nih.gov/
S-4 Effects on Bone Mineral Density
According to the study by Gao et. Al, treatment with S-4 over an 8-week course showed that there was a significant increase in bone mineral density in castrated male rats.
In a study conducted by Kearbey et. Al, female Sprague-Dawley rats were treated to see the effects of the SARM on the accelerated bone loss caused by ovariectomy. This study concluded that the S-4 treatment maintained cortical content and overall bone mineral density, as well as increased bone strength while also decreasing overall body fat (https://pubmed.ncbi.nlm.nih.
Additionally, the study conducted by Kearbey et Al. came to the overall conclusion that S-4 treatment has positive effects on skeletal muscle and bones. It was suggested the S-4 treatment could reduce instances of fracture by both increasing bone mineral density and strengthening bones, as well as strengthening skeletal muscles which reduce the probability of falls (https://pubmed.ncbi.nlm.nih.
